Mediomics Bridge-it assay的基礎概念是把蛋白質的DNA結合位置剪切成兩個DNA。每個“半結合位”都包含一個互補的單股片段區域，其用途可以再造成一個完整功能的蛋白結合位。這種實驗設計在缺乏蛋白質的情況下, 只有少部分的DNA“半結合位”會互相作用。
不同於將DNA結合位置分成二個”半結合位”的方法，二個PINCERs (binders)分別可辨認二個不同的抗原決定基，這二個PINCERs含有短訊號而且具有一對可以互相結合的螢光基團，在沒有目標物的情況下，二個PINCERs不會互相組合，直到目標物的出現，才導致二個PINCERs互相組合，進而使螢光訊號改變。
